چکیده

The ability of dopamine1 (D1) receptors to inhibit luminal Na -H exchanger (NHE) activity in renal proximal tubules and induce a natriuresis is impaired in spontaneously hypertensive rats (SHR). However, it is not clear whether the defect is at the level of the D1 receptor, Gsa, or effector proteins. The coupling of the D1 receptor to Gsa and NHE3 was studied in renal brush border membranes (BBM), devoid of cytoplasmic second messengers. D1 receptor, Gsa, and NHE3 expressions were similar in SHR and their normotensive controls, Wistar-Kyoto rats (WKY). Guanosine-59-O(3-thiotriphosphate) (GTPgS) decreased NHE activity and increased NHE3 linked with Gsa similarly in WKY and SHR, indicating normal Gsa and NHE3 regulation in SHR. However, D1 agonists increased NHE3 linked with Gsa in WKY but not in SHR, and the inhibitory effects of D1 agonists on NHE activity were less in SHR than in WKY. Moreover, GTPgS enhanced the inhibitory effect of D1 agonist on NHE activity in WKY but not in SHR, suggesting an uncoupling of the D1 receptor from Gsa /NHE3 in SHR. Similar results were obtained with the use of immortalized renal proximal tubule cells from WKY and SHR. We conclude that the defective D1 receptor function in renal proximal tubules in SHR is proximal to Gsa /effectors and presumably at the receptor level. The mechanism(s) responsible for the uncoupling of the D1 receptor from G proteins remains to be determined. Because the primary structure of the D1 receptor is not different between normotensive and hypertensive rats, differences in D1 receptor posttranslational modification are possible. Detection of apical Na(+)/H(+) exchanger activity inhibition in proximal tubules induced by acute hypertension.

We previously showed that acute arterial hypertension induces an inhibition of fluid and NaCl reabsorption in proximal tubules of Sprague-Dawley rats, which is associated with a rapid reversible internalization of apical Na(+)/H(+) exchanger in brush border. To determine whether there is a corresponding inhibition of apical Na(+)/H(+) exchanger activity in proximal tubules to account for the re...

AT2 Receptor Activation Prevents Sodium Retention and Reduces Blood Pressure in Angiotensin II-Dependent Hypertension.

RATIONALE Compound 21 (C-21) is a highly selective nonpeptide angiotensin AT2 receptor (AT2R) agonist. OBJECTIVE To test the hypothesis that chronic AT2R activation with C-21 induces natriuresis via an action at the renal proximal tubule (RPT) and lowers blood pressure (BP) in experimental angiotensin II (Ang II)-dependent hypertension. METHODS AND RESULTS In rats, Ang II infusion increased...

The Li+-Na+ exchange and Na+-K+-Cl- cotransport systems in essential hypertension.

This review examines the physiological functions of the Li+-Na+ exchanger and Na+-K+-Cl- cotransport system in human red blood cells. Both transporters are family aggregated and determined mainly by genetic factors; they are present in kidney and vascular cells, where they are regulated by vasoactive substances. To assess the physiological function of these two transporters, we investigated the...

Can we use erythrocytes for the study of the activity of the ubiquitous Na+/H+ exchanger (NHE-1) in essential hypertension?

Both Na+/Li+ countertransport and electrochemical proton gradient (delta mu(H+))-induced Na+ and H+ fluxes are increased in erythrocytes from patients with essential hypertension. It was assumed that these abnormalities are related to ubiquitous (housekeeping) forms of the Na+/H+ exchanger (NHE-1). To examine this hypothesis, we compared kinetic and regulatory properties of erythrocyte Na+/Li+ ...
